- at 10:32 AM EKKA 2006!![]() amount chocos mel bought! the bottom right fudges are mine... 2 only lah... bought other stuff as well! ![]() stars sponge! sooooo cute! how i wish i can play with them now... miss those days @ The Magic Hall... most importantly, i miss my mr pmc... - at 7:23 PM i ate a mutated strawberry!!! it was flat, fat and big! it is time to put my revision aside and relax for atinywhile.. after all the rushing of SDLs, PBLs and revision i seriously need to have a gd rest. its like a routine now to wake up at 6:45am and slp at a weird timing between 1-3am at night.. or should i say morning.. is it normal to have this type of lifestyle when u are in uni? i can feel the pressure and stress building up at the back of my head.. i think its because of that one freaky subject - Human Endocrinology... why on earth did i even choose to take this subject man? coz it was the only available subject that fits perfectly into my timetable... if i were to change it to any others, i would have to change the whole timetable... so there. side track, my toe nail polish is coming out!!! i need to get a nail polish removal soon before my toe nails turn ugly... the going-to-come-out polish will look like some dirt sticking on to the nails. anyway.. i feel quite relaxed today! hehe... completed my gene therapy SDL all thanks to mel as she chionged me to get it done before the lecturer can even ask us to do. left a bit of revision to do and that's it for today! things on hand: - ecology lit review - gonorrhoea summary (due in 3 weeks time) die! tml i must chiong for the gonorrhoea summary! arrrrrrrrrrrrrrgh...... alright i shall stop here and continue my revision... i feel like a toot toad. i am going to watch movie after revision... casablanca! - at 6:59 PM the ideal viral vector for gene therapy should be: - safe - readily made - easily introduced into the appropriate target tissues in patients - provide life-long expression of the gene of interest at appropriate levels. sadly to say, at present there is no single viral vector that can fulfil all these factors yet as they will either lack one or two of the factors. to date, there are 3 classes of viral vectors being used and each of the viral vector has their own characteristics: 1) retrovirus pros: - can accomodate up to 8kb of inserts (DNA segments), commodious enough for many genes that might be transffered - only allow low number of the DNA to integrate with the chromosome and the integrated DNA is stable - non toxic to the cells cons: - viral DNA does not integrate with non-dividing cells therefore it is unable to use in many of the tissues. 2) adenovirus pros: - able to integrate in dividing and non-dividing cells - able to accomodate inserts from 30 to 35 kbs - they can be obtained at high titer and able to infect a wide variety of cell types cons: - often elicit a strong immune and inflammatory response - they are able to multiple independently (episomal) and expressed only transiently in the target cells. 3) adeno-associated virus pros: - they are said to be the best viral vectors as they elicit no adverse reaction - they also infect dividing and non-dividing cells - they can exist episomally or stably integrated into a host chromosome. cons: - but they can only accomodate inserts up to 5kb... so there... good things comes in small package..
